ATTENUATION OF OXIDATIVE STRESS AND NEUROTOXICITY BY MK-801 (DIZOCILPINE) ON DIPENTYLPHTHALATE-INDUCED COGNITIVE DYSFUNCTION IN MICE

Objectives: The aim of our research is to study the effect dipentylphthalate (DPeP), a plasticizer on cognition and various oxidative stress markers in mice, explore modulatory effects MK-801. Methods: In present study, experimental mice were orally treated with two doses (33 100 mg/kg) DPeP for 28 days. Cognitive functions assessed using spatial navigation task Morris water maze (MWM) step-down latency (SDL) passive avoidance apparatus. Oxidative was by examining levels malondialdehyde (MDA), glutathione (GSH), ferric-reducing antioxidant power (FRAP), 8-hydroxy-deoxyguanosine (8-OH-dG) whole brain mice. Results: exposure led statistically significant increase decline SDL apparatus when compared control groups. showed following administration as seen rise MDA, 8-OH-dG, fall GSH FRAP levels. Conclusion: data suggest that could adversely affect learning memory an stress-mediated neuronal damage pre-administration MK-801 has potential attenuate these effects.

Dizocilpine (MK-801) prevents the development of sensitization to ethanol in DBA/2J mice.

DBA/2J mice repeatedly exposed to ethanol (2 g/kg, i.p. every 48 h for 8 days) exhibit sensitization to the locomotor stimulant effects of ethanol. Pretreatment with the NMDA receptor antagonist dizocilpine (0.2-0.3 mg/kg, i.p.) completely prevents the development of sensitization. Thus, NMDA receptors appear to play an important role in behavioural sensitization to ethanol.
